Abstract
The objective of this study was to obtain high-quality genomic DNA from an appropriate volume of milk and apply it to bovine breed identification and individual recognition.Briefly,6 mL of milk was centrifuged at 4000 r/min and 3~5℃for 15~25 min;the upper fat layer and middle skim layer were discarded,and the bottom pellet was retained.The pellet was then resuspended in sterile PBS buffer(pH value 7.3~7.5)to generate a resuspension,which was again centrifuged at 4000 r/min and 3~5℃for 8~12 min;the supernatant was removed,and the final pellet was collected.Genomic DNA was extracted from these cells using a magnetic-bead protocol compatible with automated platforms,thereby increasing throughput,shortening processing time,and reducing labor costs.Results showed that this method yields single-band,non-degraded,high-quality DNA from only 6 mL of milk,although yields vary among individuals due to differences in somatic cell counts.The extracted DNA meets the requirements for gene-chip genotyping and next-generation sequencing.关键词
