苍术中聚炔类化学成分及其抗菌活性
Polyynes in Atractylodis Rhizoma and Their Antibacterial Activities

Abstract
Objective:To study the chemical constituents of Atractylodis Rhizoma and evaluate their antibacterial activities.Methods:Chromatographic techniques such as silica gel column chromatography,Sephadex LH-20 gel chromatography,and semi-preparative high-performance liquid chromatography were used for separation and purification.The structures of the compounds were identified based on spectral data combined with literature data.The antibacterial activities of the compounds against Staphylococcus aureus,carbapenem-resistant Escherichia coli,and methicillin-resistant S.aureus were determined by the two-fold dilution method.Results:Four polyacetylenes were isolated from the dried rhizomes of Atractylodes lancea and identified as 2-[(1E,7Z)-nona-1,7-dien-3,5-diyn-1-yl]furan(1),atractylodin(2),atractylodinol(3),and(4E,6E,12E)-tetradeca-4,6,12-trien-8,10-diyne-1,3-diol(4).Among them,atractylodin A showed a good inhibitory effect on carbapenem-resistant E.coli,and(4E,6E,12E)-tetradeca-4,6,12-trien-8,10-diyne-1,3-diol exhibited a good inhibitory effect on methicillin-resistant S.aureus,with minimum inhibitory concentrations of 20 mg·mL-1 and 32 µg·mL-1,respectively.Conclusion:Compound 1 is a new compound,named atractylodin A,and compound 4 is isolated from the genus Atractylodes for the first time.Both compounds exhibit inhibitory activities against drug-resistant strains.In-depth study of their antibacterial mechanisms is expected to provide new insights for the research and development of improved antibacterial agents.关键词
