Abstract

Objective:To study the anti-glioma activity of morins and mulberrofurans from the flower branch of Gastrodia elata.Methods:The glioma HS683 cells were used,and the anti-glioma activity of sogastrodinol(1),gastrodinol B(2),gastrodinol C(3),isotetrapterol(4),cyclomorusinol hydroperoxide(5),benzylkuwanon C(6),and benzylmulberrofuran G(7)from the flowering branch of G.elata was investigated by cell proliferation assay(MTS),cell scratch assay,and flow cytometry-based cell cycle analysis methods.Results:Compounds 2-5 and 7 significantly inhibited the proliferation of glioma HS683 cells,with their half inhibitory concentration(IC50)values of 63.54,26.99,12.90,12.17,and 11.73 μmol·L-1,respectively.Moreover,the results of the cell scratch assay showed that the morins 4,5 and mulberrofuran 7 exhibited a substantial inhibitory effect on the migration of HS683 cells.Further cell cycle analysis revealed that compounds 4 and 7 induced a significant M phase arrest in HS683 cells,while compound 5 induced a significant G1 phase arrest in HS683 cells,thereby exerting anti-glioma activity.Conclusion:Compounds 4,5,and 7 showed the most significant anti-glioma activity,and they have the potential to be used as lead molecules.This study provides a scientific theoretical basis for the targeted development of the flowering branch of G.elata resources and anti-glioma active products,as well as drug development.
