Comparison of conception rates between fresh and frozen embryo transfer in sheep

Abstract
This study aimed to systematically compare the conception rate differences between fresh and frozen-thawed embryo transfers in sheep,and to evaluate the survival capacity of blastocysts with varying expansion degrees after cryopreservation and thawing.Blastocysts cultured in vitro to day 7 were used as materials,and recipient ewes were randomly divided into a fresh embryo transfer group and a frozen-thawed embryo transfer group.The fresh embryo group underwent direct embryo transfer,while the frozen embryo group involved blastocysts preserved by programmed freezing methods and transferred after thawing.Simultaneously,blastocysts were graded based on their expansion degree prior to freezing,and the survival rates of different grades after thawing were compared.All embryos were transferred into the uterus of synchronized recipient ewes via laparoscopy,and the transplantation outcomes across different sheep breeds were further analyzed.The results showed that the survival rates of Grade A(fully expanded)and Grade B(expanded)blastocysts were(96.36±1.23)%and(93.93±2.11)%,respectively,significantly greater than that of Grade C(less expanded)blastocysts(P<0.01),indicating a strong correlation between blastocyst expansion degree and post-thaw viability.The overall conception rate of the frozen-thawed embryo transfer group was 63.88%,slightly greater than the 61.26%of the fresh embryo transfer group,with no significant difference(P>0.05).However,the frozen-thawed group demonstrated favorable pregnancy outcomes across multiple breeds,highlighting its practical applicability.The findings suggest that,with mature cryopreservation and thawing techniques,frozen-thawed embryo transplantation can achieve conception rates comparable to fresh embryos,while offering advantages such as long-term preservation and convenient transportation,making it suitable for industrial applications.关键词
