Abstract

Objective:To investigate the mechanism of Lysimachia christinae Hance(LCH)in inhibiting cholesterol stone formation.Methods:Forty C57BL/6J mice aged six weeks were divided randomly into four groups:control group,model group,high-dose group and low-dose group.Except for the control group,mice in other groups were fed by a high-cholesterol lithogenic-inducing diet to build the model.The low-dose group and high-dose group were gavaged with 20 mg/kg and 80 mg/kg of LCH extract,meanwhile the mice in control group and model group were gavaged with the equal amount of physiological saline.The experiment lasted for eight weeks.Key evaluation indicators included cholesterol stone formation rate,weight,and serum levels of total cholesterol(TC),triglycerides(TG),total bile acids(TBA),low-density lipoprotein cholesterol(LDL-C),and high-density lipoprotein cholesterol(HDL-C)were evaluated in each group.Transcriptomics sequencing was performed to analyze liver differentially expressed genes(DEGs)and functional enrichment analysis.The mRNA expression levels of key DEGs(FXR,SHP,CYP7A1,CYP27A1,BSEP)were validated by quantitative real-time PCR(qRT-PCR).Results:Compared with the control group,the model group exhibited significantly higher gallstone formation rate,cholesterol stone weight,and levels of TC,TG,and LDL-C(P<0.01),indicated successful model establishment.LCH treatment dose-dependent reduced these parameters mentioned(P<0.01)while significantly increased TBA levels in bile(P<0.01),suggested that LCH promotes the conversion of cholesterol to bile acids.Transcriptome sequencing revealed 82 significantly upregulated and 78 significantly downregulated genes in the LCH-treated groups compared with the model group.KEGG enrichment analysis indicated that the DEGs were primarily associated with pathways related to bile acid metabolism and bile acid excretion.According to the qRT-PCR result,LCH treatment group significantly upregulated the mRNA expression of FXR,SHP,CYP27A1,and BSEP compared with the model group(P<0.01).Conclusion:By activating hepatic FXR,promoting cholesterol conversion to bile acids via the alternative pathway and enhances bile acid transport into the bile canaliculi,Lysimachia christinae Hance can reduce cholesterol levels and inhibit cholesterol stone formation.
