A One-Step Detection Platform for Pertussis Based on Loop-Mediated Isothermal Amplification and Enzyme-Free Nucleic Acid Circuits

Abstract
Respiratory infections are one of the most common types of diseases,among which pertussis is an acute respiratory infection caused by Bordetella pertussis(BP),which is highly contagious and has a long course.Infants and young children,due to their weak immunity,may develop serious complications and even may be life-threatening when infected with pertussis.In this study,a one-step detection platform that integrated loop-mediated isothermal amplification(LAMP)with enzyme-free nucleic acid circuit(catalytic hairpin assembly,CHA),termed as LAMP-CHA,was developed.The target gene was exponentially amplified through LAMP,and the amplicons were subsequently recognized by CHA to generate fluorescence signals.The LAMP-CHA platform enabled detection of as low as 1 copy/μL of the conserved gene of BP,exhibiting high sensitivity and specificity.Application to clinical samples further confirmed its reliability and practicality,underscoring its potential as an efficient and accurate tool for clinical diagnosis.关键词
