Simultaneous Detection of Aflatoxin B1 and Ochratoxin A in Grains Using A Magnetic Separation Dual Color Afterglow Luminescence Aptasensor

Abstract
Food security is of great importance to national economic development and public health.In this work,a magnetic separation dual color persistent luminescent aptasensor(Apt-PLNPs@cDNA-Fe3O4)was developed to meet the requirements of highly sensitive detection of the co-contamination of aflatoxin B1(AFB1)and ochratoxin A(OTA)in grains.The sensor was constructed by integrating adapter functionalized afterglow luminescent nanoparticles(Apt-PLNPs)with complementary chain modified Fe3O4 magnetic particles(cDNA-Fe3O4).The target molecules AFB1 and OTA were competitively bound to their respective aptamers(Apt1 and Apt2)in the sensor,forming AFB1-Apt1-Zn2GeO4∶Mn2+(ZGM)and OTA-Apt2-Zn1.25Ga1.5Ge0.25O4∶Cr3+,Yb3+,Er3+(ZGGOC)complexes,which promoted the dissociation of cDNA1-Fe3O4 and cDNA2-Fe3O4.This method enabled the persistent luminescent nanoparticle ZGM to release corresponding afterglow luminescence signals at a wavelength of 535 nm and ZGGOC at a wavelength of 700 nm,respectively,thereby achieving rapid,background free,and highly specific quantitative detection of AFB1 and OTA mycotoxin.The experimental results showed that the afterglow luminescence intensity of ZGM and ZGGOC had a good linear relationship with concentrations of AFB1 and OTA in the range of 0.001‒50 ng/mL,with detection limits of 3.4×10-4 and 3.7×10-4 ng/mL,respectively.The prepared Apt-PLNPs@cDNA-Fe3O4 sensor was applied to detection of AFB1 and OTA contents in grain samples,with recoveries from 93.5%to 101.2%.This study provided an innovative and effective detection strategy for early warning of mycotoxin contamination in grains.关键词
