栉孔扇贝内脏多糖的提取分离与生物活性研究
Extraction,isolation,and bioactivity of visceral polysaccha-rides from Chlamys farreri

Abstract
In this study,we examined the extraction,separation,and activity of polysaccharides from Chlamys far-reri viscera byproducts.The crude polysaccharides were primarily prepared using the enzyme digestion method.The optimal enzyme digestion conditions were identified using a one-way experiment,which revealed an optimal neutral protease to trypsin ratio of 1∶2 at pH 7,a reaction temperature of 40℃,and a digestion time of 2 h.The crude polysaccharides were subjected to separation and purification using a DEAE-52 ion exchange column and a Sephadex G-100 gel exclusion chromatographic column,respectively.The molecular weight of the purified CFP4-G3 fraction was 4 935 Da,with a uronic acid content of 4.081%and a sulfate content of 4.335%.Infrared absorption spectroscopy and ultraviolet spectroscopy confirmed that CFP4-G3 conformed to the typical structural features of amino polysaccharides.Bioactivity assays of the resulting CFP4-G3 polysaccharide demonstrated that it had strong antioxidant activity,including scavenging of hydroxyl radicals,superoxide anion,and DPPH,and also possessed substantial reducing power.In addition,CFP4-G3 exhibited strong immunostimulatory activity.Moreover,CFP4-G3 substantially stimulated NO release and the secretion of TNF-α and IL-6 in RAW264.7 mouse macro-phages.The above findings provide a crucial theoretical foundation for the in-depth development of scallop viscera polysaccharides.关键词
