摘要

Abstract

The aim of this study was to analyze the effect of Toll-like receptor 7/8(TLR7/8)activator R848 on sperm function in dairy cows and to preliminarily investigate its mechanism of action.After co-in-cubation of Holstein cow semen with R848,sperm motility performance and sperm quality parameters were analyzed and the expression levels of fatty acid β-oxidation(FAO)-related proteins were detected by Western blot.The results showed that after co-incubation with R848,the proportion of spermatozoa that swam to the upper layer was significantly lower than that of the control group(P<0.05),and the kinetic parameters of the lower spermatozoa(VSL,VCL,VAP)were significantly lower than that of the control group and the upper spermatozoa(P<0.05),whereas there was no significant difference between sperm viability and acrosomal and plasma membrane intact rates(P>0.05).Western blot results showed that the expression levels of IKBA and ACC proteins in the spermatozoa of dairy cows in the R848 co-incubation group were significantly higher,whereas the expression level of CPT1 protein was significantly lower com-pared with the control group(P<0.05).The results showed that the TLR7/8 activator R848 could inhibit cow's sperm viability without affecting sperm function by reducing cow sperm ATP production through the IKBA/ACC/CPT1/FAO pathway.
