Ultrasonic-assisted Ionic Liquid Extraction of Total Flavonoids from Nanzhang Cornus Officinalis and Their Antioxidant Activity

Abstract
Objective:To extract total flavonoids from Nanzhang Cornus Officinalis and study their antioxidant activity.Methods:Total flavonoids were extracted by ultrasonic-assisted ionic liquid extraction.With the yield of total flavonoids as the indicator,or-thogonal design was carried out on the basis of single factor test to optimize the prescription and process.The morphological changes of the medicinal materials before and after extraction were characterized,and the antioxidant activity of the extract was investigated by DPPH and OH free radical scavenging experiments.Results:The optimal process conditions for ultrasonic-assisted ionic liquid extraction of total flavonoids from Nanzhang Cornus Officinalis are as follows:the extraction solvent is 1.0 mol/L 1-hexyl-3-methylimidazolium tetrafluoroborate([C6mim]BF4),the solid-liquid ratio is 1:8,the ultrasound time is 30 min,and the ultrasonic power is 320 W.Under these conditions,the yield of total flavonoids from Cornus Officinalis was 12.51%,and the morphology of the extracted Cornus Officinalis medicinal material underwent significant changes.The IC50 values of total flavonoids from Cornus Of-ficinalis for scavenging DPPH and OH free radicals were 0.335 mg/mL and 0.487 mg/mL,respectively.Conclusion:Ultrasonic-as-sisted ionic liquid extraction of flavonoids from Nanzhang Cornus Officinalis is an effective and feasible extraction method.The ex-tracted total flavonoids from Nanzhang Cornus Officinalis have good antioxidant activity,providing a scientific experimental basis for their development and utilization.关键词
