Abstract
Objective To investigate the neuroprotective and mechanism of monotropein(Mon)on the N2a cell infected by adenoviral carrying the APPswe gene.Methods The CCK-8 assay was used to detect the optimal concentration by Mon and Nrf2 activator(Nrf2-Ac)on APP-N2a cells,flow cytometry was used to detect the effect of Mon and Nrf2-Ac on apoptosis of APP-N2a cells,ELISA assay was used to detect the effect of Mon and Nrf2-Ac on inflam-cytokines of APP-N2a cells and immunohistochemistry assay was used to detect the effect of Mon and Nrf2-Ac on Nrf2,HO-1 protein expression of APP-N2a cells.Results The results showed that 20 μM Mon and 1000 nM Nrf2-Ac could increase the survival rate and reduce apoptosis of the APP-N2a cells.The levels of IL-6,IL-1β,TNF-α were decreased while the level of Nrf2,HO-1 protein expression was increased.Conclusion Mon has the most significant neuroprotective in the APP-N2a cells by activating the Nrf2/HO-1 signaling pathway.关键词
