Effects of Songyu Anshen Prescription on Melatonin Synthesis and Receptor Expression in a Rat Model of Insomnia

Abstract

Objective:To evaluate whether Songyu Anshen Prescription(SYF)modulates melatonin synthesis and melatonin re-ceptor expression in rats with p-chlorophenylalanine(PCPA)-induced insomnia.Methods:Specific pathogen-free(SPF)male Sprague Dawley(SD)rats were assigned to a control group(n=8)and a model cohort(n=40)rendered insomniac by intraperito-neal PCPA(IP).After model establishment,rats were randomized into five groups(n=8 each):Model(normal saline intragastric gavage,IG),SYF(17 g/kg IG),Luzindole(Luz;1 mg/kg IP),SYF(17 g/kg IG)+Luz(1 mg/kg IP),and Agomelatine(2.25 mg/kg IG).Treatments were administered once daily for 7 days;the control and model groups received an equal volume of normal saline.Serum melatonin was measured by enzyme-linked immunosorbent assay(ELISA).Arylalkylamine N-acetyltrans-ferase(AANAT)in the pineal gland and hypothalamic melatonin receptor 1B(MTNR1B)gene were quantified by western blot-ting.Results:Relative to controls,model rats exhibited significantly lower serum melatonin and reduced protein expression of pine-al AANAT and hypothalamic MTNR1B(P<0.01).Versus the model group,both the SYF and Agomelatine groups significantly increased serum melatonin and up-regulated pineal AANAT and hypothalamic MTNR1B(P<0.01).Compared with SYF,Luzin-dole alone and SYF plus Luzindole significantly lowered these measures(P<0.01),indicating attenuation of SYF's effects by me-latonin receptor antagonism.Conclusion:SYF exhibits melatonin receptor agonist-like activity in PCPA-induced insomnia rats.Its mechanism may involve upregulating pineal AANAT to enhance melatonin synthesis and engaging hypothalamic melatonin recep-tor-mediated signaling.
