热带假丝酵母细胞膜通透性改造对十二碳二元酸合成效率的影响
Modifications of the cell membrane permeability of Candida tropicalis and the impacts on the efficiency of dodecanedioic acid synthesis

Abstract
Dodecanedioic acid(DDA),an important long-chain di-carboxylic acid,has extensive industrial application prospects and significant synthetic value.Despite the potential use in DDA-production,C.tropicalis strain has limited efficiency for DDA synthesis due to the issue of cell membrane permeability for hydrophobic substrates such as dodecane.Ergosterol is a key component of fungal cell membrane,and the blockage of its synthesis can significantly alter the cell membrane properties.Herein,homologous recombination technology was utilized in our studies to knock out the genes encoding C-5 sterol desaturase(ERG3)and 14 α-demethylase(ERG11),two important enzymes involved in the lanosterol biosynthesis pathway in C.tropicalis.Upon the optimization of fermentation conditions,the synthesis efficiency for DDA was greatly improved.The experimental results showed that the cell membrane permeability was significantly enhanced for the knockout strains.By comparison with the non-knockout strain,the DDA yields of both ERG3-deficient strain and ERG11-deficient strain increased by 38.50%and 19.33%,respectively.Further optimization of the shake flask fermentation conditions led to 9.03 g/L yield of DDA after the treatment for 144 h.Our research work provides a new idea for regulating cell membrane permeability,i.e.,through genetic engineering to improve the metabolic efficiency of hydrophobic substrates,and offers a theoretical basis for the application of C.tropicalis in biomanufacturing.关键词
