Characteristic Analysis of Specific Neutralizing Antibody Subtype in Serum of Children Infected with Norovirus

Abstract
Objective To understand the characteristics of serum antibody subtypes,associated cytokine changes,and cross-neutralizing activity in children infected with Norovirus(NoV)can provide valuable references for the development of NoV antibodies and vaccines.Methods A total of 27 children infected with NoV who were admitted to the Children's Hospital in Kunming from April to July 2025 were selected as the infection group,while 2 healthy children during the same period served as the control group.ELISA was used to detect the binding titers of serum samples from the infection group against the prevalent NoV strains GⅡ.4 and GⅡ.17,as well as the antiserum antibody subtypes,the area under the curve(AUC)of the dose-response curve was employed to evaluate serum titers and antibody subtype characteristics.An in vitro surrogate neutralization assay was conducted to detect the neutralizing activity of serum antibodies,with the 50%blocking titer(BT50)used to represent the ability to block virus-receptor binding.Flow cytometry was applied to measure the types and levels of cytokines in the infection group and the control group serum samples.The normality of cytokine levels and serum antibody subtypes was assessed using the Shapiro-Wilk test,and correlation analysis was performed using Spearman's method.Results The majority of serum samples were capable of binding to GⅡ.4/GⅡ.17 NoV VLPs,with GⅡ.4 being the dominant type.Analysis of the AUC of antigen-specific antibody subtypes revealed that in over 96%of the infected samples,the binding strength of IgA and IgM was higher than that of IgG,with IgM exhibiting the most intense response.In the in vitro surrogate neutralization assay,8 samples targeting GⅡ.4 VLPs reached the positive detection threshold for BT50,whereas the neutralizing titers against GⅡ.17 VLPs were generally low,with only 3 samples testing positive.Additionally,no significant differences were observed in cytokine levels between the infection group and the control group(t=0.25~1.41,all P>0.05).However,within the infection group,a positive correlation was identified between IL-6 and IL-10 expression levels(r=0.55,P<0.05).Meanwhile,IFN-γ levels showed positive correlations with GⅡ.4 VLP-specific IgM,IgA,IgG,and GⅡ.17 VLP-specific IgA levels(r=0.38~0.59,all P<0.05).Similarly,IL-10 levels were positively correlated with both GⅡ.4 and GⅡ.17 VLP-specific IgA and IgG levels(r=0.48~0.57,all P<0.05).Conclusions The specific antibodies mediating the blocking effect in the sera of children infected with NoV are primarily of the IgA and IgM subtypes.The cytokine expression profiles in these patients indicate a dynamic equilibrium between pro-inflammatory and anti-inflammatory factors.Furthermore,certain correlations are observed between cytokine levels and antibody subtypes.关键词
