Experimental Study of the Effect of TIM-3 Silencing-Regulated TAMs Polar-ization on the Proliferation and Migration of Cervical Cancer Cells

Abstract
Objective This study aim to investigate the regulatory impact of the T cell immunoglobulin and mucin domain-con-taining protein-3(TIM-3)gene silencing on the polarization of tumor-associated macrophages(TAMs)and its influence on the proliferation and migration of cervical cancer cells.Methods The experiment constructed a TAMs model with silenced TIM-3 gene,and used CCK-8 assay to detect the proliferation ability of cervical cancer cells,Transwell assay to detect the migration abil-ity of cervical cancer cells,Western blotting to detect the expression changes of epithelial-to-mesenchymal transition(EMT)-related proteins(E-cadherin,N-cadherin),and flow cytometry to detect the polarization state of TAMs.Results The proliferation and migration rates of cervical cancer cells with TIM-3 silencing were significantly decreased,and the differences were statistically significant(t= 4.17~12.81,all P<0.05).The protein expression level of E-cadherin was significantly increased,while that of N-cadherin was significantly decreased,and the differences were statistically significant(t=8.90~25.00,all P<0.05).The propor-tion of CD86-positive cells increased significantly,while the proportion of CD163-positive cells decreased significantly,and the differences were statistically significant(t=11.92~26.10,all P<0.05).Conclusions The TIM-3 gene silencing regulates effec-tively the polarization of tumor-associated macrophages(TAMs),thereby suppressing the proliferation and migration of cervical cancer cells.This finding provides a new theoretical foundation and a promising therapeutic target for the immunotherapy of cer-vical cancer.关键词
