黑木耳多糖提取工艺优化及其体外抗氧化与抗糖活性研究
Study on the optimization of the extraction process of polysaccharide from Auricularia auricula and its antioxidant and hypoglycaemic activity in vitro

Abstract
This study employed enzymatic hydrolysis to extract polysaccharides from Auricu-laria auricula.The extraction conditions for the polysaccharides were optimized through sin-gle-factor experiments and response surface methodology,using polysaccharide yield as the primary indicator.Two polysaccharide components,designated AAP-1 and AAP-2,were isolated via DEAE cellulose column chromatography,and their total sugar,protein,uronic acid,and total phe-nol contents were quantified.The antioxidant capacities of AAP-1 and AAP-2 were evaluated based on their scavenging rates of DPPH radicals,hydroxyl radicals,and superoxide anions.Additionally,the hypoglycemic effects of AAP-1 and AAP-2 were assessed through their inhibition rates on α-am-ylase and α-glucosidase.The results revealed that the optimal extraction conditions for polysaccha-rides from Auricularia auricula are as follows:enzymatic hydrolysis pH of 5,temperature of 50 ℃,duration of 90 minutes,and enzyme concentration of 40 mg/g.Compared to crude polysaccharides,the total sugar content in the eluted AAP-1 and AAP-2 significantly increased,while impurities such as protein,uronic acid,and total phenols were markedly reduced.Specifically,at a concentration of 0.8 mg/mL,AAP-1 and AAP-2 demonstrated DPPH radical scavenging rates of 49.05%and 62.69%,hydroxyl radical scavenging rates of 34.04%and 45.02%,and superoxide anion scaven-ging rates of 38.62%and 39.98%,respectively.Furthermore,at a concentration of 1.0 mg/mL,the inhibition rates of AAP-1 and AAP-2 on α-glucosidase were 72.24%and 87.10%,respectively,while their inhibition rates on α-amylase were 80.51%and 89.23%,respectively.These findings in-dicate that both AAP-1 and AAP-2 possess notable antioxidant and hypoglycemic properties,with AAP-2 displaying superior in vitro activity,thereby providing a theoretical foundation for the further development of polysaccharides derived from Auricularia auricula.关键词
