摘要

Abstract

In situ hybridization(ISH),a pivotal cytogenetic technique,plays a critical role in deciphering plant species origins,resolving phy-logenetic relationships between cultivated crops and their wild relatives,tracing parental chromosomal contributions in interspecific hybrids,conducting karyotype analyses and characterizing meiotic chromosome behavior.Based on probe-labeling strategies,ISH is categorized into ge-nomic in situ hybridization(GISH)and fluorescence in situ hybridization(FISH).Maize(Zea mays),as the largest crop in China,has ex-perienced a significant decline in genetic diversity due to domestication and selective breeding.Maintaining and enhancing the genetic diversi-ty of corn is crucial for sustained genetic gains in breeding.Wild relatives of maize,such as teosintes and Tripsacum L.,possess rich genetic variation and exhibit stronger tolerance to abiotic and biotic stresses.The review synthesizes recent advances in ISH applications for elucida-ting phylogenetic relationships,karyotype evolution,and chromosomal composition in maize and its wild relatives.Furthermore,we highlight how the integration of next-generation sequencing data with oligonucleotide(Oligos)-based probe design-targeting species-specific repetitive sequences,chromosomal regions,or whole chromosomes-enables precise cytogenetic markers.Notably,combinatorial FISH platforms incorpora-ting multiple repeat probes are poised to accelerate research in maize cytogenetics,genome evolution,and the exploitation of elite alleles from wild germplasms.As genomic resources for maize and its relatives become increasingly comprehensive,these refined ISH methodologies will provide robust technical frameworks for advancing both fundamental and applied studies in maize improvement.
