摘要

Abstract

The family of glycoside hydrolase 12 is a class of enzymes widely existing in nature,which plays a key role in biological processes such as cellulose degradation,xylan degradation,plant cell wall metabolism,pathogen infection and symbiotic relationship.The three-dimen-sional structure of this family of enzymes is centered on the β-folded barrel,with highly conserved active sites and substrate binding grooves,which determines their specific hydrolysis ability to substrates and shows a certain diversity in different biological sources.GH12 family en-zymes are catalyzed by a retention mechanism,which has been fully verified by crystal structure elucidation,mutation experiments and compu-tational simulations.At present,the crystal structures of dozens of enzymes in GH12 family members have been elucidated,which provides an important basis for in-depth study of their structure-function relationships.In addition,Loops and key amino acid residues in the enzyme structure play an important role in the catalytic process,which may affect the substrate specificity,catalytic efficiency and stability of the en-zyme.In this paper,the members and structure of the GH12 family,the function of enzymes,the location and mechanism of enzyme activity,and the study of GH12 in important plant pathogenic filamentous fungi are re viewed,and based on these research results,the study of GH12 function in different fungi,the analysis of the catalytic mechanism of GH12 enzyme activity,and the search for GH12 interacting proteins are prospected.
