摘要

Abstract

[Objective]β-ketoacyl-ACP synthase Ⅲ(KAS Ⅲ),as the core initiating enzyme in the plant fatty acid synthesis pathway,plays a key role in regulating oil accumulation and fatty acid synthesis.By analyzing the expression patterns and functional characteristics of the Co-KAS Ⅲ gene family in different tissues and under various stress conditions in Camellia oleifera,the present study aims to provide a theoretical foundation for further elucidating the biological functions of CoKAS Ⅲ genes.[Method]Members of the CoKAS Ⅲ gene family in hexaploid C.oleifera were systematically identified using bioinformatics methods,followed by comprehensive analyses of their gene structures,evolution-ary relationships and expression profiles.[Result]A total of 15 CoKAS Ⅲ gene family members were identified,all containing the PLN02326 conserved domain specific to the KAS Ⅲ family.Members within the same subfamily exhibited highly consistent gene structures and function-al domains,suggesting functional similarity.Physicochemical property analysis revealed significant diversity among family members,and sub-cellular localization predictions indicated that these proteins were primarily distributed in the chloroplast(3),mitochondria(4),plasma membrane(5),organelle membrane(1)and extracellular space(1).Phylogenetic analysis classified CoKAS Ⅲ into four subfamilies,with subfamilies Ⅰ and Ⅱ clustering with homologous proteins from other plants,demonstrating evolutionary conservation,while subfamilies Ⅲ andⅣ were unique to hexaploid C.oleifera,indicating gene family expansion driven by polyploidization.Collinearity analysis elucidated the genet-ic relationships among C.oleifera,Arabidopsis thaliana,C.sinensis and C.vietnamensis.Promoter analysis identified numerous cis-acting ele-ments related to light response,hormonal regulation and stress response.Expression pattern analysis showed that A2.8G267000,C2.7G18850 and C2.13G163200 were specifically highly expressed in flower buds,while most genes were active in the early stages of seed development.C2.13G107700 was significantly expressed in both susceptible(SH)and resistant(RH)varieties.C1.13G106400 was markedly up-regula-ted in response to low-temperature stress(LS04,2 ℃).B2.11G177300 and B1.11G165900 were specifically expressed under short-day(SD)and long-day(LD)conditions,respectively,whereas B1.5G165800,B2.1G358800 and C2.7G188500 were specifically expressed un-der medium-day(MD)conditions.During seed germination,expression levels exhibited fluctuating changes.[Conclusion]The study identi-fies 15 CoKAS Ⅲ gene family members,which exhibit both evolutionary conservation and specificity.Differential expression patterns across tissues,anthracnose resistance,low-temperature stress and photoperiod responses suggest that this gene family plays important roles in the growth and development,disease resistance and adaptation to abiotic stresses in C.oleifera.
