Abstract

[Objective]The present paper aimed to identify MIKCC-type MADS-box genes in Vernicia montana and explore superior haplo-types involved in the reproductive organ development regulatory network.[Method]Bioinformatics methods were used to analyze sequence characteristics,evolutionary relationships,conserved domains,chromosome localization,promoter elements and gene expression profiles of MIKCC-type MADS-box gene haplotypes at the whole-genome scale in V.montana.A yeast two-hybrid assay was conducted to study protein functional differentiation.[Result]A total of 31 MIKCC-type MADS-box genes were identified,among which 5 genes had amino acid se-quence haplotype differences,and 13 genes had cis-acting element haplotype differences.Analysis of promoter elements and gene expression suggested that MIKCC-type MADS-box genes in V.montana may broadly participate in floral organ development,floral transition and seed de-velopment through hormonal regulatory pathways.Among these genes,VmMADS3.1,VmMADS32.2 and VmMADS48.2 were identified as su-perior haplotypes.VmMADS3.1 may be associated with the delayed developmental timing of female flower formation in V.montana;Vm-MADS32.2 likely promoted stamen differentiation while suppressing carpel development through a dosage-dependent regulatory mechanism;whereas VmMADS48.2 may promote abortion of stamens in female flowers,and play critical roles in regulating ovule formation,seed size and morphological patterning.[Conclusion]VmMADS3.1,VmMADS32.2 and VmMADS48.2 were identified as superior haplotypes involved in the reproductive organ development regulatory network in V.montana.The study provides theoretical foundation for future application of MIKCC-type MADS-box genes in regulating sex differentiation and improving seed traits in V.montana.
