化橘红药材来源植物化州柚组织培养技术研究
Study of tissue culture technology for Citrus grandis 'Tomentosa',the plant source of H uajuhong

Abstract
Huajuhong is a traditional Chinese medicine.and Citrus grandis'Tomentosa'is one of its plant sources.To establish a complete tissue culture technology system for C.grandis'Tomentosa',young stem segments of C.grandis Tomentosa'were used as explants to investigate the effects of different influencing factors on bud induction,proliferation,seedling strengthening,rooting culture as well as transplant survival.The results showed that the optimal disinfection method for stem segment explants was a 30-second immersion in 75%ethanol,followed by sterilization with 0.1%mercuric chloride(HgCl2)for 6-7 minutes.For primary culture,the optimal culture medium for axillary bud induction was MS+1.5 mg/L 6-BA+1.0 mg/L KT+0.5 mg/L NAA.For adventitious bud proliferation subculture,a suitable medium was MS+1.0 mg/L 6-BA+0.5 mg/L NAA.The optimal medium for seedling strengthening and rooting was 1/2 MS+1.0 mg/L IBA+0.5 mg/L NAA+1 g/L activated carbon.The best transplant substrate for tissue-cultured rooted seedlings was a mix of peat soil∶sheep manure soil∶perlite at a volume ratio of 2∶1∶1,which resulted in robust roots and a survival rate of over 75%.The established rapid propagation system through stem segment tissue culture of C.grandis'Tomentosa'can provide technical reference for large-scale seedling production and lay a theoretical foundation for its genetic transformation research.关键词
