Screening of SRAP Core Primers for Sugar Beet Based on Touchdown Program

Abstract
[Objective]The main objective is to select SRAP core primers with high polymorphism and high stability for the identification of genetic diversity in sugar beet.[Methods]Amplification was performed using SRAP primer combinations under a Touchdown program.The products were separated by 8%non-denaturing polyacrylamide gel electrophoresis,and primers meeting the criteria were selected based on data analysis of the amplification results.[Results]From 504 pairs consisting of 24 upstream primers and 21 downstream primers,43 pairs of primer combinations with high polymorphism and stability were identified,of which 40 pairs of primer combinations had a polymorphism information content(PIC)value greater than 0.75,24 pairs of primer combinations had a PIC value greater than 0.8,and 4 pairs of primer combinations had a PIC value greater than 0.9.The PIC range was 0.657 to 0.953,and the average PIC value of primer pairs was 0.812.[Conclusion]The screening of SRAP core primers provides theoretical support for the identification of genetic diversity in sugar beet,as well as for the selection of parents for further cross-breeding and molecular marker-assisted breeding.关键词
