Research progress of pyroptosis in rheumatic immune diseases
摘要
Abstract
Pyroptosis is a form of programmed cell death characterized by its pro-inflammatory nature,mediated by activated proteins from the gasdermin family.This process is typically marked by the formation of permeable pores in the plasma membrane,leading to gradual cellular swelling and eventual lysis.Pyroptosis is accompanied by the indiscriminate release of inflammatory mediators,such as interleukin-1β(IL-1β)and interleukin-18(IL-18),along with other intracellular contents.Pyroptosis plays a crucial role in maintaining tissue homeostasis and defending against pathogens by modulating immune defense and inflammatory responses.However,excessive or aberrant activation of pyroptosis may result in pathological inflammation and contribute to the pathogenesis of rheumatic diseases to varying extents.This review aims to provide a comprehensive overview of the historical evolution of the definition of pyroptosis,its characteristics,the activated molecular mechanisms,and the recent advancements in pyroptosis research related to rheumatic disorders,aiming to open new perspectives for mechanistic studies and future targeted therapies of these diseases.关键词
