基于AMPK/mTOR通路探讨异维A酸软胶囊联合火针疗法对痤疮模型的影响
Effects of Isotretinoin Soft Capsule Combined with Fire Needle Therapy on Acne Model Based on AMPK/mTOR Pathway

Abstract
Objective In order to explore the effects of isotretinoin soft capsule combined with fire needle therapy on acne model,and to analyze its potential mechanism based on AMP-activated protein kinase(AMPK)/mammalian target of rapamycin(mTOR)pathway.Methods Totally 60 mice were randomly divided into six group(n=10):control group,acne model group,isotretinoin group,fire needle group,combined treatment group and inhibitor group.A mouse acne model was constructed by intradermal injection of 50μL propionibacterium acnes suspension on the back.The back skin of the mice was observed,and the area of acne was measured.Hematoxylin-eosin(HE)staining was used to observe the pathological changes of skin lesions.Oil Red O staining and Nile Red fluorescence staining were used to evaluate the level of lipid accumulation in skin lesions.Western blotting was used to analyze the expression levels of AMPK/mTOR pathway and autophagy-related proteins.Results Compared with the control group,the acne model group showed a significant increasing in acne area,Oil Red O staining area,Nile Red staining fluorescence intensity,and the expression level of phosphorylated mammalian target of rapamycin(p-mTOR)/mTOR,while the expression levels of phosphorylated adenosine monophosphate-activated protein kinase(p-AMPK)/AMPK,Beclin-1,autophagy-related gene 7(ATG7),and the ratio of microtubule-associated protein 1 light chain 3-Ⅱ(LC3-Ⅱ)to LC3-Ⅰ were significantly decreased(P<0.05).Compared with the isotretinoin group and the fire needle group,the acne area,Oil Red O staining area,Nile Red staining fluorescence intensity and p-mTOR/mTOR expression level of the combined treatment group were significantly reduced,and the expression levels of p-AMPK/AMPK,Beclin1,ATG7,and LC3-Ⅱ/LC3-Ⅰ were significantly increased(P<0.05).Compared with the combined treatment group,the acne area,Oil Red O staining area,Nile Red staining fluorescence intensity and p-mTOR/mTOR expression level in the inhibitor group were significantly increased,and the expression levels of p-AMPK/AMPK,Beclin1,ATG7 and LC3-Ⅱ/LC3-Ⅰ were significantly decreased(P<0.05).Conclusion Isovitamin A acid soft capsule combined with fire needle therapy can significantly improve acne lesions in mice and inhibit the secretion of sebaceous glands.The mechanism may be achieved by activating autophagy mediated by AMPK/mTOR signaling pathway.关键词
