lncRNA DLEU1在皮肤黑色素瘤中的ceRNA调控机制研究
ceRNA Regulatory Mechanism of lncRNA DLEU1 in Cutaneous Melanoma
摘要
Abstract
Objective In order to investigate the expression and regulatory mechanism of long non-coding ribonucleic acid deleted in lymphocytic leukemia 1(lncRNA DLEU1)in skin cutaneous melanoma(SKCM).Methods Clinical samples from 8 SKCM patients after surgery were collected to detect the expression of lncRNA DLEU1.The subcellular localization of lncRNA DLEU1 was detected by fluorescence in situ hybridization(FISH).The downstream targets of lncRNA DLEU1 were predicted using the StarBase 3.0 online tool(https://starbase.sysu.edu.cn/).Small interfering RNA(siRNA)was used to knock down lncRNA DLEU1 in A375 cells.Immunohistochemistry was used to detect protein expression levels,and quantitative real-time polymerase chain reaction(RT-qPCR)was used to detect gene expression levels.Scratch assay and transwell assay were used to detect cell migration and invasion.Results The lncRNA DLEU1 was localized in the cytoplasm.According to StarBase 3.0 predictions,the downstream targets of lncRNA DLEU1 were miR-126-5p and growth factor receptor-bound protein 2-associated binding protein 1(GAB1).In clinical samples,lncRNA DLEU1(both P=0.002)and GAB1(P=0.001)were overexpressed,while miR-126-5p was underexpressed(P=0.001)in cancer tissues compared to adjacent non-cancerous tissues.Immunohistochemistry also showed high expression of GAB1 in cancer tissues.In A375 cells,siRNA was used to knock down lncRNA DLEU1,resulting in significantly reduced expression of lncRNA GAB1(P=0.016)and significantly increased expression of miR-126-5p(P=0.003)in the knockdown group.Knockdown of lncRNA DLEU1 significantly reduced the migration,invasion and proliferation of A375 cells.Conclusion The lncRNA DLEU1/miR-126-5p/GAB1 network is closely related to the occurrence and development of SKCM,and is expected to become a new target for the diagnosis and treatment of SKCM.关键词
