摘要

Abstract

Objective In order to investigate the effects of application frequency and dosage of exogenous olive oil on the clinical severity of seborrheic dermatitis(SD)and its correlation by the cyclooxygenase-2(COX-2)/prostaglandin E2(PGE2)/prostaglandin E receptor(EP)signaling pathway.Methods A total of 50 SD guinea pig models were divided into five groups using the simple random ranking method:Group A(0.5 mL applied daily),B(1.0 mL applied daily),C(1.0 mL applied every 3.5 days),D(0.5 mL applied every 3.5 days)and E(model control),with 10 animals in each group.Experimental groups were received topical olive oil for two weeks.Intervention efficacy was evaluated through clinical scoring,histopathological assessment and molecular analyses.The pathological changes were observed by hematoxylin-eosin(HE)staining.The protein expressions of COX-2 and PGE2 were detected by enzyme-linked immunosorbent assay(ELISA).Immunohistochemical(IHC)staining was performed to determine the expression profiles of EP subtypes.Results Group B exhibited the highest clinical severity scores compared to group A,C and D(P<0.01,P<0.001,P<0.01).The score of group C was lower than that of group E(P<0.05).Group B showed marked epidermal hyperkeratosis,acanthosis and inflammatory cells infiltration.The expression levels of COX-2 and EP2 in group A and B were higher than those in group D(P<0.05,P<0.01),while the expression level of EP3 in group B was lower than that in group D(P<0.05).The expression level of EP4 in group B was higher than that in group E(P<0.05).There was no statistically significant difference in PGE2 and EPl between the groups(P>0.05).Conclusion Daily topical application of high-dose olive oil(1.0 mL applied daily)may exacerbate SD by upregulating COX-2,EP2 and EP4 expression,which promotes pro-inflammatory responses,coupled with insufficient EP3 expression that attenuates anti-inflammatory effects.
