朱琏兴奋Ⅰ型针法对糖尿病神经源性膀胱大鼠氧化应激的影响
Effect of ZHU Lian's Excitatory Type Ⅰ Acupuncture on Oxidative Stress in DNB Rats

Abstract
Objective:To observe the effects of ZHU Lian's excitatory type Ⅰ Acupuncture on oxidative stress-related indicators in rats with diabetic neurogenic bladder(DNB)and to explore its potential mechanisms in treatment of DNB.Methods:50 SD rats were randomly divided into the control group,the model group,the Western medication group,the conventional acupuncture group,and the study group,with 10 rats in each group.The DNB model was established by intraperitoneal injection of streptozotocin(STZ).The Western medication group was intervened with intragastric administration of Methylcobalamin,the conventional acupuncture group was treated by needling Zhongji(RN3),Sanyinjiao(SP6),Lieque(LU7),and Taichong(LR3)with a mild reinforcing-reducing technique,and the study group was given ZHU Lian's excitatory typeⅠ acupuncture at the same acupoints.Bladder wet weight was measured in each group.HE staining was used to observe morphological changes in bladder tissue.Western blot was performed to detect expressions of MDA and SOD.Expression of iNOS was measured by ELISA,and RT-PCR was used to detect mRNA expressions of SOD and iNOS.Results:Compared with the control group,the model group showed a significant increase in bladder wet weight(P<0.01),as well as elevated expressions of MDA,iNOS,and iNOS mRNA(P<0.01),whereas expressions of SOD and SOD mRNA were significantly decreased(P<0.01).Compared with the model group,the Western medication group,the conventional acupuncture group,and the study group all demonstrated significantly reduced wet bladder weights and decreased expressions of iNOS and iNOS mRNA(P<0.01),along with lower MDA levels(P<0.05),and increased expressions of SOD and SOD mRNA(P<0.01).Compared with the conventional acupuncture group,both the Western medication group and the study group exhibited lower wet bladder weight and MDA levels(P<0.05),decreased iNOS and iNOS mRNA expressions(P<0.01),and higher SOD and SOD mRNA expressions(P<0.05,P<0.01).There were no statistical differences in wet bladder weights,expressions of MDA,SOD,SOD mRNA,iNOS,or iNOS mRNA between the Western medication group and the study group(P>0.05).Conclusion:Oxidative stress plays a key role in occurrence and development of DNB in rats.ZHU Lian's excitatory type Ⅰ acupuncture exerts a protective effect on bladder function in diabetic rats.Its mechanism may be related to reducing lipid peroxidation,enhancing antioxidant enzyme activity,and regulating expressions of related proteins and mRNAs to alleviate oxidative stress-induced damage.This may be one of the underlying mechanisms by which this acupuncture method treats DNB.关键词
