Effects of Jiaji EA on Programmed Necrosis-Related Factors in Rats with SCI

Abstract
Objective:To investigate the effects of Jiaji electro-acupuncture(EA)on programmed necrosis-related factors in the spinal cord tissue of rats with spinal cord injury(SCI),and to clarify the mechanism by which Jiaji EA promotes the repair of spinal cord tissue damage.Methods:Allen's SCI model was established using an NYU Impactor M-Ⅲ.36 SD rats were randomly divided into the sham operation group,the model group,and the Jiaji EA group,with 12 rats in each group.Each group was further subdivided into 3-day subgroup(n=6)and 7-day subgroup(n=6).In the 7-day subgroup,BBB scores were assessed at 6 h,1 d,3 d,and 7 d to evaluate hindlimb motor function.In both 3-day and 7-day subgroups,serum samples were collected after anesthesia for ELISA analysis and injured spinal cord tissues were collected for HE staining and qRT-PCR detection.The expressions of programmed necrosis-related factors(RIPK1,RIPK3,MLKL)and necrosis-related inflammatory mediators(HMGB1,IL-6)were analyzed.Results:BBB score analysis showed that the Jiaji EA group had significantly higher scores on both 3 d and 7 d compared with the model group(P<0.05).HE staining revealed that,relative to the model group,the Jiaji EA group exhibited fewer necrotic cavities in the spinal cord,reduced neuronal swelling,more preserved gray matter structure,less neuronal loss,and alleviated inflammatory cell infiltration.qRT-PCR results showed that on 3 d and 7 d after surgery,the mRNA expressions of programmed necrosis-related factors(RIPK1,RIPK3,MLKL)in spinal cord tissue were significantly higher in both the model group and the Jiaji EA group compared with the sham operation group(P<0.01).The Jiaji EA group showed significantly lower mRNA expressions of RIPK1,RIPK3,and MLKL compared with the model group after the treatment(P<0.05).Comparisons between the 7-day and 3-day model subgroups revealed significant differences in mRNA expressions of RIPK3 and MLKL(P<0.05),and similar significant differences were observed between the 7-day and 3-day Jiaji EA subgroups(P<0.05).ELISA results showed that the expressions of HMGB1 and IL-6 were significantly higher in the model group than in the sham operation group(P<0.01).The Jiaji EA group showed significantly reduced expressions of HMGB1 and IL-6 compared with the model group after the treatment(P<0.05).Significant differences in the expressions of HMGB1 and IL-6 were observed between the 7-day and 3-day subgroups in both the model group and the Jiaji EA group(P<0.05).Conclusion:Following SCI,necrosis of tissue cells leads to the release of inflammatory factors,with a marked accumulation at 3 days post-injury and a decline by day 7.Jiaji EA has been found to inhibit spinal cord cell necrosis and the release of inflammatory mediators,thereby protecting spinal cord tissue and promoting repair of the injury.关键词
