Abstract

In order to clarify the resistance levels of different tomato varieties to bacterial canker,iden-tify resistant tomato varieties,and elucidate the underlying resistance mechanisms,a needle-puncture inoculation method was used to evaluate the resistance of 66 tomato varieties to this disease.The activi-ties of antioxidant enzymes,malondialdehyde(MDA)content,and the expression levels of disease resistance-related genes were measured in highly resistant and highly susceptible tomato varieties after inoculation with Clavibacter michiganensis,the pathogen causing tomato bacterial canker.The results showed that the resistance of the 66 tomato varieties to bacterial canker could be classified into five grades:Micro Tom was highly resistant,Red Brandy resistant,23 varieties including Fushan 99,Bail-ingqianxi,and Xiaoqianjinhuangfei moderately resistant,27 varieties including Provence,Niuxin Tomato,and Heimeiren susceptible,and 14 varieties including Yellow Pear,Micai Tomato,and Hezuo 909 highly susceptible.Based on the manifestation of resistance,the tested tomato varieties were divided into three types:21 varieties were resistant to pathogen invasion,12 varieties resistant to patho-gen expansion,and 33 varieties dual-sensitive.The activities of peroxidase(POD),superoxide dis-mutase(SOD),and catalase(CAT)in the highly resistant variety Micro Tom inoculated with C.michi-ganensis were significantly higher than those in the highly susceptible variety Hezuo 909.At 72 hours post-inoculation,the POD and CAT activities of the highly resistant variety were 2.28 times and 1.40 times those of the highly susceptible variety,respectively.At 120 hours post-inoculation,the SOD activity of the highly resistant variety was 1.72 times that of the highly susceptible variety,while the MDA content of the highly resistant variety was significantly lower than that of the highly susceptible variety.Real-time quantitative RCR analysis indicated that infection by C.michiganensis induced sig-nificant upregulation of the pathogenesis-related protein-encoding genes LePOD and PR-NP24 in the highly resistant variety.These findings demonstrate that the resistance of tomato varieties to C.michi-ganensis is closely associated with their antioxidant capacity,membrane lipid peroxidation level,and the expression levels of pathogenesis-related genes.
