肥胖相关基因FTO表达水平与高脂血症性急性胰腺炎的相关性分析
Correlation Analysis of the Expression Level of the Fat Mass and Obesity as Sociated Gene FTO and Hypertriglyceridemic Pancreatitis
摘要
Abstract
Objective:To analyze the correlation between the expression level of obesity-related genes(FTO)and hyperlipidemic induced acute pancreatitis(HTGP).Method:A total of 100 patients with HTGP admitted to the Second Affiliated Hospital of Fujian Medical University from January 2023 to December 2024 were selected and divided into the mild group(n=65)and the moderate to severe group(n=35)according to the severity grading diagnostic criteria of acute pancreatitis(AP)in the"Chinese Guidelines for the Diagnosis and Treatment of Acute Pancreatitis(2021)".Another 50 patients with hyperlipidemia without AP who were admitted during the same period were selected as the control group, and 30 healthy individuals undergoing physical examinations were selected as the healthy group. The differences in the expression levels of triglycerides and FTO mRNA among the four groups were compared. The Pearson correlation coefficient was used to analyze the correlation between the expression level of FTO mRNA and the triglyceride level.Multivariate logistic regression was used to analyze the relationship between the expression level of FTO mRNA and the severity of HTGP.The receiver operating characteristic(ROC)curve was drawn to evaluate the predictive efficacy of FTO mRNA expression level for the severity of HTGP.Result:The body mass index(BMI),the proportion of alcohol consumption history, and the expression levels of triglycerides and FTO mRNA in the moderately severe group, mild group, and control group were all higher than those in the healthy group, and the differences were statistically significant(P<0.05).The expression levels of BMI,triglycerides, and FTO mRNA in the moderately severe group and mild group were higher than those in the control group, moderately severe group>mild group>control group, and the differences were statistically significant(P<0.05).Pearson correlation analysis showed that the expression level of FTO mRNA was positively correlated with the triglyceride level(r=0.447,P<0.001).Univariate logistic regression analysis showed that BMI,history of alcohol consumption, triglyceride level, and FTO mRNA expression level were all related to the severity of HTGP(P<0.05).Multivariate logistic regression analysis showed that BMI,triglyceride, and FTO mRNA expression levels were independent risk factors for the severity of HTGP(P<0.05).ROC curve analysis showed that the areas under the ROC curves(AUC)of BMI,triglycerides, and FTO mRNA expression levels for predicting the severity of HTGP were 0.791,0.847,and 0.868,respectively.The predictive efficacy of FTO mRNA was superior to that of BMI(Z=2.968,1.996,P<0.05).When the cut-off value of 0.40 was taken, the sensitivity and specificity of FTO mRNA in predicting the severity of HTGP were 90.00%and 76.80%,respectively.Conclusion:FTO mRNA is highly expressed in patients with HTGP,which is a risk factor affecting the severity of HTGP and has a good predictive value for the severity of HTGP.关键词
