基于脂质组学探讨何首乌致特异质肝损伤的潜在机制
Lipidomics Reveals Potential Mechanism of Heshouwu-Induced Idiosyncratic Liver Injury

Abstract
Objective:To explore the synergistic effect of Heshouwu(何首乌)and the susceptible factor tumor necrosis factor(TNF)-α on liver injury and its effect on lipid metabolism.Methods;C57 mice were randomly allocated into blank,Heshouwu(3.12 g/kg and 6.24 g/kg),TNF-α(10 ng/g),TNF-α(10 ng/g)+Heshouwu(3.12 g/kg or 6.24 g/kg)groups.The liver tissue and plasma samples of mice in each group were collected,and the histopathological changes were observed.The levels of liver injury markers alanine aminotransferase(ALT),as-partate aminotransferase(AST),alkaline phosphatase(ALP),direct bilirubin(DBIL)and pro-inflammatory cytokines interleukin(IL)-6 and IL-1β in the plasma were determined.Plasma lipidomics based on ultra-high performance liquid chromatography-mass spectrometry was employed to analyze changes in lipid metabolism.Results:Compared with the blank group,the TNF-α+Heshouwu(3.12 g/kg)and TNF-α+Heshouwu(6.24 g/kg)groups showed expansion of the portal area and obvious inflammatory infiltration in the liver tissue and raised levels of ALT,AST,ALP,IL-6,and IL-1β(P<0.05,P<0.01).Lipidomics results showed that compared with the blank group,TNF-α+Heshouwu(6.24 g/kg)led to obvious abnormalities in lipid metabolism,which were manifested as elevated levels of phosphatidylcholine,phosphatidyle-thanolamine,phosphatidylglycerol,phosphatidylserine,partial triglycerides,diglycerides,sphingomyelin,ceramide,hexaglycosylceramide,zy-mosterol,and coenzyme.Conclusion:Heshouwu combined with TNF-α can lead to liver injury in mice by regulating lipid metabolism and ag-gravating inflammatory responses.关键词
