电针通过调控RCAN1促进脑缺血小鼠血管新生与运动功能恢复
Electroacupuncture Promotes Angiogenesis and Motor Functional Recovery in Mice with Cerebra Ischemia by Regulating Regulator of Calcineurin 1

Abstract
Objective To observe the effect of electroacupuncture at Baihui and Dazhui points on the expression of regulator of calcineurin 1(RCAN1)in the peri-infarct area of a mouse model of cerebral ischemia,and to investigate whether RCAN1 medi-ates the role of electroacupuncture in promoting angiogenesis and improving motor function.Methods Male C57BL/6 mice were randomly divided into normal group,model group,electroacupuncture group,and knockdown+electroacupuncture group,with 6 mice in each group.The method of photochemical thrombosis was used to establish a model of cerebral ischemia.The pole test and the front paw grip strength test were used to assess motor function;TTC staining was used to measure in-farct area;quantitative real-time PCR and Western blot were used to measure the mRNA and protein expression levels of RCAN1;super-resolution microscopy was used to measure vascular density and ratio;ELISA was used to measure the levels of the inflammatory factors interleukin-1β(IL-1β)and interleukin-6(IL-6).Results Compared with the normal group,the model group had significant increases in pole climbing time and retention time(P<0.05),a significant reduction in front paw grip score(P<0.05),a significant increase in infarct area(P<0.05),significant reductions in vascular density and ratio(P<0.05),significant increases in the mRNA and protein expression levels of RCAN1(P<0.05),and significant increases in the levels of IL-1β and IL-6 in the peri-infarct area(P<0.05).Compared with the model group,the electroacupuncture group had significant improvements in the above indicators(P<0.05),and after RCAN1 knockdown,the effect of electroacupuncture was significantly weakened(P<0.05).Conclusion Electroacupuncture improves motor functional recovery in mice with cere-bral ischemia by upregulating the expression of RCAN1 in the peri-infarct area,reducing infarct area,increasing vascular densi-ty,and inhibiting inflammatory response.关键词
