Abstract

Saline and alkaline soils in Xinjiang region severely limit the growth of alfalfa(Medicago sativa)and the development of the alfalfa industry.In this study,'Xinmu No.4'was used to compare the mechanism of saline and alkaline stress inhibition of alfalfa seed germination By setting different concentration gradients(0,25,50,100 mmol·L-1)of NaCl and NaHCO3 solutions as well as solution Ⅰ[50 mmol·L-1 NaCl(pH 6.37)];solution Ⅱ[50 mmol·L-1 NaHCO3(pH 8.10)],solution Ⅲ[50 mmol·L-1 NaCl+NH3·H2O(pH 8.10)],and solution Ⅳ[50 mmol·L-1 NaHCO3+HCl(pH 6.37)]stress treatments,to compare mechanisms of action of saline and alkaline stress inhibition of alfalfa seed germinations.The results showed that saline stress significantly inhibited alfalfa seed germination but NaHCO3 inhibited alfalfa seed germination to a greater extent.Salt stress inhibited the amount and rate of water uptake throughout sprouting but alkali stress promoted the amount and rate of water uptake in the fast and slow stages of seed water uptake,but inhibited water uptake in the growth water uptake stage.In addition,increasing the pH of the NaCl solution did not exacerbate the degree of inhibition of seed germination,whereas decreasing the pH of the NaHCO3 solution did not alleviate the degree of inhibition of seed germination.In conclusion,salt stress affected alfalfa seed germination by inhibiting alfalfa seed water uptake as well as later embryonic root and germ growth.Alkali stress promoted swelling and water absorption during the early stages of alfalfa seed germination,and significantly inhibited physiological metabolism within the seed as well as the growth of the radicle and germ after seed bleaching.The difference in the effects of saline and alkaline stress on alfalfa seed germination was not caused by pH effects alone.
