Abstract

To enhance the exploration of high-quality native grass germplasm resources in Tibet,this study comprehensively evaluated the cold resistance of 21 wild Elymus germplasm resources collected from different regions of Tibet,along with the domesticated variety E.sibiricus'Chuancao 2'.After germination,the seedlings were transferred to trays filled with vermiculite and regularly irrigated with a 1/2 Hoagland nutrient solution.After 30 d,the seedlings were randomly subjected to 4℃for 48 h to induce cold stress,while those kept at 24℃served as the control.The results showed that,compared with the control,cold stress significantly increased(P<0.05)the content of malondialdehyde(MDA)in the leaves of seven germplasm lines,activity of superoxide dismutase(SOD)in 14 germplasm lines,peroxidase activity of peroxidase(POD)in eight germplasm lines,proline(Pro)in 16 germplasm lines,initial fluorescence Fo in nine germplasm lines,and non-regulatory energy dissipation[Y(NO)]of PSⅡ in six germplasm lines.In contrast,the maximum fluorescence Fm of the three germplasm lines,photochemical quenching coefficient qP of seven germplasm lines,actual photochemical efficiency[Y(Ⅱ)]of 10 germplasm lines,photosynthetic electron transfer rate(ETR)of 12 germplasm lines,and maximum light energy conversion efficiency(Fv/Fm)of PSⅡ in the three germplasm lines significantly decreased(P<0.05).The results indicate abundant genetic variation in the collected germplasm resources possessed.A comprehensive evaluation using a weighted membership function method revealed that germplasm entries 16,17,23,4,and 27 exhibited strong cold tolerance,and strengthening their research,development,and utilization is recommended.
