独活寄生汤对兔膝关节软骨损伤的修复作用及机制研究
Reparative effects and mechanisms of Duhuo Jisheng Decoction on knee articular cartilage injury in rabbits

Abstract
Objective To investigate the effects and mechanisms of Duhuo Jisheng Decoction(DHJSD)on the repair of knee cartilage injury in rabbits.Methods Twenty white rabbits were randomly divided into five groups(n=4 each),including sham surgery group,model group,low-dose DHJSD group,medium-dose DHJSD group,and high-dose DHJSD group.A rabbit knee cartilage injury model was established using a ring drill.The sham surgery group and model group received physiological saline by gavage,while the DHJSD groups received low,medium,or high doses of DHJSD by gavage for 12 weeks.At week 12,the Mankin score was used to evaluate knee cartilage injury.Quantitative reverse transcription PCR(qRT-PCR)was used to detect type Ⅱ collagen mRNA expression in the repair tissue at the cartilage injury site.ELISA was used to measure joint fluid levels of ADAMTS-5 and TGF-β1.Immunohistochemistry was used to observe p-Smad3 expression in the repair tissue.Results Compared with the sham surgery group,the model group had significantly higher Mankin scores(P<0.05).Compared with the model group,all DHJSD groups had significantly lower Mankin scores(P<0.05),and the medium-and high-dose groups had lower scores than the low-dose group(P<0.05).Type Ⅱ collagen mRNA expression at the injury site was significantly lower in the model group than in the sham surgery group(P<0.05),and was significantly higher in the medium-dose DHJSD group compared with the model group(P<0.055).Compared with the sham surgery group,the model group had increased ADAMTS-5 and decreased TGF-β1 levels in joint fluid(P<0.05).Compared with the model group,the medium-and high-dose DHJSD groups showed decreased ADAMTS-5 and increased TGF-β1 levels(P<0.05).Compared with the low-dose group,medium-and high-dose DHJSD groups showed decreased ADAMTS-5 and increased TGF-β1 levels(P<0.05).Immunohistochemistry showed uniform p-Smad3 expression in the sham group.The model group showed little p-Smad3 expression at the cartilage injury site.The low-dose group showed sparse expression,while medium-and high-dose groups showed pronounced p-Smad3 expression at the repair site.Conclusion Duhuo Jisheng Decoction promotes repair of knee cartilage injury and reduces inflammatory response in joint fluid.Its mechanism may be related to activation of the TGF-β/Smad signaling pathway.关键词
