摘要

Abstract

Objective To investigate the protective effects and mechanisms of Huashi Runzao Huoxue Formula(HRHF)against concanavalin A(ConA)-induced autoimmune hepatitis(AIH)in mice.Methods Forty mice were randomly divided into five groups(n=8 per group),including blank group,model group,RIP1 inhibitor(Nec-1)group,low-dose HRHF group,and high-dose HRHF group.The blank group received 15 mg/kg physiological saline via tail vein injection,while the other groups were injected with 15 mg/kg ConA to establish the AIH model.Two hours after modeling,mice in the blank and model groups were given 10 mL/kg saline by gavage;the low-and high-dose HRHF groups received 1.95 g/kg and 3.90 g/kg HRHF by gavage,respectively;the Nec-1 group received 1 mg/kg Nec-1 by intraperitoneal injection.All treatments were administered once daily for 10 consecutive days.Liver and spleen morphology,liver and spleen indices,liver histopathology,serum liver function indicators,liver inflammatory and oxidative stress markers,and mRNA and protein expression of the RIP1/RIP3/MLKL signaling pathway were assessed.Results Compared with the blank group,the model group showed increased liver and spleen indices(P<0.05);liver histopathology revealed congestion,necrosis,and extensive inflammatory cell infiltration;serum liver enzymes and bilirubin were elevated;albumin was decreased(P<0.05);hepatic inflammatory and oxidative stress markers were increased(P<0.05);and mRNA and phosphorylated protein levels of RIP1,RIP3,and MLKL were upregulated(P<0.05).Compared with the model group,mice in the low-and high-dose HRHF groups and the Nec-1 group had decreased liver and spleen indices(P<0.05),improved liver histopathology,reduced serum liver enzyme levels,decreased hepatic inflammatory and oxidative stress markers(P<0.05),and downregulated mRNA and phosphorylated protein expression of RIP1,RIP3,and MLKL(P<0.05).Conclusion HRHF can significantly attenuate liver injury in ConA-induced AIH mice,and its mechanism may be related to inhibition of programmed necrosis via blocking the oxidative stress-induced RIP1/RIP3/MLKL signaling pathway.
