不同时间间隔传代对牦牛瘤胃厌氧真菌群落多样性的影响
Effects of time interval sampling on the diversity of the rumen anaerobic fungal community in yak

Abstract
[Objective]To investigate the effects of different passage time intervals on the diversity of anaerobic fungi in the yak rumen.[Method]Anaerobic fungi were isolated from yak rumen chyme using a consecutive batch culture series with intervals of 3 days(D3),5 days(D5),and 7 days(D7),repeated seven times.Fungal mycelium was collected,and isolates were identified through ITS sequencing.[Result]The number of operational taxonomic units(OTUs),Shannon-Wiener diversity index,and Chao1 richness index of anaerobic fungi increased progressively with longer passage intervals.A Wilcoxon rank-sum test revealed significant differences in fungal abundance be-tween D5 vs.D3 and D7 vs.D3(P<0.05),but not between D7 vs.D5(P>0.05).At the phylum level,the identi-fied fungi primarily belonged to Basidiomycota,Ascomycota,Mortierellomycota;The dominant orders were Walle-miales,Eurotiales,and Filobasidiales,while the dominant genera were Wallemia,Filobasidium,and Cladosporium.Fungi across all passage intervals were predominantly saprophytic.However,with longer intervals,the relative abun-dance of symbiotic trophic and pathogenic trophic increased.[Conclusion]Extending the passage time interval en-hances the abundance and diversity of anaerobic fungi in the yak rumen.It also shifts the fungal trophic mode from a primarily saprophytic state toward an increased presence of symbiotic and pathogenic types.关键词
