基于定量蛋白质组学探讨补肾益精方治疗2型糖尿病骨质疏松症的作用机制
To explore the mechanism of Bushen Yijing Recipe in the treatment of osteoporosis in type 2 diabetes mellitus based on quantitative proteomics

Abstract
Objective To explore the possible pharmacological mechanism of Bushen Yijing Recipe in the treatment of type 2 diabetic osteoporosis(T2DOP)based on proteomics.Methods A total of 60 male wistar rats of SPF grade were randomly selected 10 as the blank group,and the remaining 50 rats were used to construct the T2DOP rat model by intraperitoneal injection of streptozotocin(STZ)according to Tang Hui's method.The rats conforming to the T2DOP model were randomly divided into blank group,positive control group,low,medium and high dose groups of Bushen Yijing Recipe.They were given normal saline,alendronate sodium and Bushen Yijing Recipe by gavage for 8 weeks.During the period,the general condition of rats,blood glucose and body weight changes were recorded.After the intervention,the femur of the rats was scanned by Micro-CT,and the cancellous bone was reconstructed by 3D reconstruction and parameter analysis.HE was used to observe the morphological changes of rat femur.According to the results of Micro-CT and HE,a group of Bushen Yijing Recipe was selected to compare with the model group and the blank group by the method of stable isotope dimethylation labeling.Quantitative proteomics technology was used to explore the protein network and mechanism of Bushen Yijing Recipe in the intervention of T2DOP.Differential proteins were screened and verified by Western Blot.Results(1)According to the results of Micro-CT scanning 3D reconstruction and HE staining,Bushen Yijing Recipe can improve the bone metabolism balance,improve bone microstructure and enhance bone mechanical properties of T2DOP rat model.(2)Through proteomics,107 differential proteins(82 up-regulated proteins and 24 down-regulated proteins)were screened between the blank group and the T2DOP model group.Compared with the T2DOP model group,78 differentially expressed proteins(15 up-regulated proteins and 63 down-regulated proteins)were screened in the high-dose group.GO and KEGG analysis and PPI network analysis were performed on the screened differentially expressed proteins.It was found that there were interactions between 23 differentially expressed proteins,forming a complex regulatory network.The core proteins in the network included MMP9,TNF,Akt1.These core proteins are at the hub of the PPI network.Conclusion Bushen Yijing Recipe can inhibit bone resorption and promote bone formation through the expression of core gene proteins such as MMP9,TNF and Akt1,so as to achieve the purpose of preventing and treating T2DOP.关键词
