Abstract

Objective Exploration of the estrogen-like effects and underlying molecular mechanisms of Siwu Decoction in regulating immune function based on network pharmacology,molecular docking,and experimental validation in animals.Methods The intersection targets related to Siwu Decoction,estrogen,and immunity were screened using databases such as TCMSP,CTD,OMIM,and ImmPort.A PPI network was constructed via the STRING online platform.GO and KEGG enrichment analyses were performed using the DAVID 6.8 database.Molecular docking was conducted using AutoDock Vina.A total of 40 BALB/c mice were randomly divided into five groups:control,model,estradiol,high-dose Siwu Decoction,and low-dose Siwu Decoction.Except for the control group,all other groups received intraperitoneal injections of cyclophosphamide[80 mg/(kg·day)]on days 1~3 of the experiment.The estradiol group received oral administration of estradiol at[0.22 mg/(kg·day)]for 7 consecutive days.The high-dose and low-dose Siwu Decoction groups were administered 20 mg/(g·day)and 10 mg/(g·day),respectively,for 7 consecutive days.Body weight change rate,spleen and thymus indices were measured.Hematoxylin and eosin(HE)staining was used to examine spleen tissue morphology.Flow cytometry was used to detect changes in CD4+/CD8+T cells and B lymphocytes in spleen tissue.RT-PCR was conducted to measure mRNA expression levels of ERβ,TNF-α,RELA,and IL-1β in spleen tissue.Immunofluorescence was used to detect protein expression levels of ERβ,RELA,and phosphorylated RELA(p-RELA)in spleen tissue.Results A total of 276 targets related to Siwu Decoction,estrogen,and immunity were identified,involving 177 signaling pathways.Molecular docking confirmed that key active components(catechin,ligustrazine,chlorogenic acid,gallic acid,and kaempferol)showed strong binding affinity with core targets(TNF,STAT3,EGFR,RELA,IL-6,and IL-1β).Siwu Decoction was found to protect immune organs and cells in mice,significantly increasing the CD4+/CD8+ratio and the number of CD19+B lymphocytes compared to the model group(P<0.05).It also promoted the expression of TNF-α,IL-1β,ERβ,RELA,and p-RELA in spleen tissue(P<0.05).Conclusion Siwu Decoction exerts estrogen-like effects in regulating immune function by mediating ERβ to activate the NF-κB signaling pathway.
