拟穴青蟹BAMBI基因的序列特征及表达分析
Sequence characterization and expression analysis of BAMBI gene in Scylla paramamosain

Abstract
In order to explore the effect of BAMBI[bone morphogenetic protein(BMP)and activin membrane-bound inhibitor]on ovarian maturation of Scylla paramamosain,we studied the expression patterns of different tissues,the temporal and spatial expression patterns of ovarian and hepatopancreas tissues at different stages of ovarian development,and the regulation of ovarian tissues in vitro treated with follicle stimulating hormone(FSH)of BAMBI.The cDNA sequence of BAMBI in S.paramamosain(Sp-BAMBI)was obtained by ovarian transcriptome data and analyzed by bioinformatics technology;the tissue specificity of Sp-BAMBI and the gene expression patterns in ovary and hepatopancreas at different stages of ovarian development were analyzed by RT-qPCR;the regulatory influence of FSH on Sp-BAMBI was investigated through in vitro culture of steroid-induced ovarian tissues from S.paramamosain.The results showed that the cDNA sequence length of BAMBI was 2 840 bp,and the open reading frame(ORF)was 984 bp in length encoding 327 amino acids.In addition,the amino acid sequence of BAMBI consisted of a highly conserved N-terminal functional domain and a highly conserved C-terminal functional domain.The phylogenetic tree analysis based on BAMBI amino acid sequences of different species showed that S.paramamosain had the closest evolutionary relationship with S.olivacea.By RT-qPCR analysis,it showed that Sp-BAMBI was expressed in all 8 tissues of S.paramamosain,including muscle,thoracic ganglion,ovary,epidermis,gill,hepatopancreas,heart and midgut,and the highest expression of Sp-BAMBI was in muscle and followed by thoracic ganglion and ovary.The Sp-BAMBI expression patterns in ovary and hepatopancreas were significantly different at different stages of ovarian development in S.paramamosain.In ovarian tissues of S.paramamosain,the expression levels of Sp-BAMBI were significantly down-regulated in stages I to II of ovarian development,significantly up-regulated in stages III to IV,and significantly down-regulated in stage V(P<0.05).Furthermore,in hepatopancreas tissues of S.paramamosain,the expression levels of Sp-BAMBI were the highest in stage I,then significantly decreased in the other 4 stages(P<0.05).Moreover,the expressions of Sp-BAMBI were significantly different from 0.1 to 10 μmol·L-1 levels in the in vitro culture of ovarian tissues induced by follicle stimulating hormone(FSH)compared with the control group in S.paramamosain(P<0.05),and 0.5 μmol·L-1 of FSH was the optimal concentration to up-regulate the gene.Thus,different concentrations of FSH regulated the expression of Sp-BAMBI significantly.The study had shown that Sp-BAMBI protein was conserved in sequence structure,which may play an important role in the regulation of oocyte development and ovarian maturation of S.paramamosain influenced by the hormone FSH.The results provided a preliminary reference for the subsequent study on the regulatory mechanism of Sp-BAMBI in ovarian development and maturation of S.paramamosain.This study also contributes to offering new insights into a model of the potential application of BAMBI-direct or indirect regulation of ovarian development in female S.paramamosain.关键词
