Abstract

This study investigated the effects of different salinity acclimation modes[(the control group A,salinity 5 ‰;slow salinity increase groups:group B(+2.5‰·d-1),group C(+5‰·d-1);fast salinity increase groups:group D(+12.5‰·d-1),group E(+25‰·d-1)]on blood physiological and biochemical parameters,antioxidant enzyme activities,and gill histology in Takifugu obscurus.Serum cortisol level,lactate dehydrogenase(LDH)activity,superoxide dismutase(SOD)activity,catalase(CAT)activity,and gill filament Na+,K+-ATPase activity were measured at 0,6,12,24,48,96,168 h after the target salinity of 30‰ was reached.The results revealed a clear dose-and time-dependent relationship between the acclimation rate and the magnitude of physiological disruption.The results of gill histology showed that compared with the control group,the degree of gill tissue damage gradually increased with the daily salinity increase rate.At 168 h,the Na+,K+-ATPase activity in the gill filaments of group B was higher than that of the other groups(P<0.05),while there was no significant difference among the other groups(P>0.05).In fast salinity increase groups,the cortisol content reached its peak at the beginning of the stress(0 h),then rapidly declined and remained at a relatively low level.In contrast,in slow salinity-increase groups,the peak of cortisol content occurred at 6 h,which was only 0.8 and 1.2 times that of the control group.This indicated that the slow salinity-increase treatment could mitigate the impact of salinity changes on T.obscurus.The LDH activity exhibited a fluctuating trend with the extension of the acclimation time.After 48 h,the LDH activity in each treatment group was significantly higher than that in the control group,and there were significant differences among the treatment groups at 168 h(P<0.05).The SOD activity in each treatment group was significantly lower than that of the control group at 0 h and 6 h(P<0.05).The SOD activity in the slow-salinity increase groups showed no significant difference from the control group after 48 h(P>0.05).There was no significant difference among all groups at 168 h(P>0.05),indicating that under slow salinity changes,T.obscurus could gradually adapt to the changes in salinity within 48 h.However,rapid salinity stress caused damage to the body's antioxidant enzyme system,and it could only gradually adapt to the salinity changes after 168 h.And high daily salinity increase inhibited CAT activity obviously,leading to the inability of the body to carry out effective antioxidant regulation.In conclusion,an excessively rapid increase in salinity can cause severe stress responses in T.obscurus,leading to an imbalance in their antioxidant system and irreversible damage to the structure of their gill tissues.However,under the acclimation mode where the daily salinity does not exceed 5‰,T.obscurus can gradually adapt to the high-salt environment for aquaculture.The results can provide a reference for the large-scale marine aquaculture of T.obscurus.
