Abstract

Body color plays an important role in species growth,reproduction and information exchanges.And it is determined by the type,distribution,quantity and pigment content of pigment cells deposited in fish.Larimichthys crocea is favored by consumers due to its high nutritional value and golden body color.Wild Larimichthys crocea generally has light yellow on the back,and golden yellow on the abdomen,lips and tail fin.The body color of farmed Larimichthys crocea degenerates.The back is light black,and the abdomen and tail fins are orange.With the continuous expansion of market demand for Larimichthys crocea and the development of Larimichthys crocea farming technology,farmed Larimichthys crocea has become the main consumption target and has great commercial values.Body color is one of the most important indicators for judging the health and freshness of Larimichthys crocea.Therefore,understanding the formation and change mechanism of the body color of Larimichthys crocea is important.In this study,farmed Larimichthys crocea was used as the research subject.The color in dorsal,abdomen,and tail of Larimichthys crocea was measured by a colorimeter.A microscope was used to analyze the composition and distribution of the pigment cells in Larimichthys crocea.A UV spectrophotometer and a liquid chromatography-tandem mass spectrometry instrument were used to analyze the content and composition of carotenoid in Larimichthys crocea.The relationship between body color parameters and carotenoid content,xanthophore characteristics,and pigment composition.All statistical analyses of the data were performed using SPSS 26.0 software.Image j was used for the quantification of xanthophores,and Origin 2021 was used for graphing.Pearson correlation analysis was chosen for relationship analysis.A total of three types of pigment cells were observed in Larimichthys crocea,namely melanophores,xanthophores,and iridophores.The dorsal skin contained considerable melanophores,and xanthophores were small and densely distributed in dorsal skin.While the abdomen skin contained a large amount of larger xanthophores with synapses.Melanophores and xanthophores were distributed in the scales.Carotenoids were predominantly deposited in the abdomen skin of Larimichthys crocea.A total of 13 carotenoids were detected in the abdomen skin,including 11 xanthophylls and 2 carotenes.Xanthophylls were the main pigments in Larimichthys crocea,with a content of(126.41±27.89))μg·g-1 and accounting for 94.89%of the total carotenoids.Among the xanthophylls.The yellow-blue value was positively correlated with the carotenoid content(P<0.05).In summary,the body color of Larimichthys crocea is related to the type,distribution,size,and pigment content of the pigment cells.This study provides a scientific basis for the further investigation of the body color formation in Larimichthys crocea.
