摘要

Abstract

[Objective]The development of cassava(Manihot esculenta)production is facing with many challenges,and viral diseases are one of the key factors limiting its ability to achieve high and stable yields,among which the prevalence of cassava common mosaic virus(CsCMV)poses a serious threat to the sustainable development of the industry.meGRXC3 is a cassava-specific CC-type glutoxin Reducing protein(GRXs),which are involved in the regulation of redox homeostasis.Previous studies have shown that TGBp1(Triple gene block 1)protein is a key factor in CsCMV pathogenesis and is involved in the regulation of host immune response.This study aims to investigate whether TGBp1 synergizes with cassava MeGRXC3 in regulating redox homeostasis in virus infestation.[Method]This study conducted a systematic validation of the mechanism of interactions between CsCMV TGBp1 proteins and MeGRXC3 by integrating yeast two-hybrid(Y2H),bimolecular fluorescence complementation(BiFC),and subcellular localization techniques.[Result]Y2H experiments confirmed that MeGRXC3 and CsCMV TGBp1 have stable mutual interaction properties in yeast system under strict screening conditions.Confocal microscopy showed that the BiFC vector of MeGRXC3 and CsCMV TGBp1 showed green fluorescence at 488 nm excitation light after co-expression in tobacco epidermal cells.Subcellular localization revealed that CsCMV TGBp1 showed co-localization in tobacco epidermal cells after co-expression with MeGRXC3,further confirming the interaction between CsCMV TGBp1 and MeGRXC3.[Conclusion]This study systematically explored the possible interactions between the CsCMV TGBp1 protein and MeGRXC3.This multidimensional research strategy provides an important experimental basis for elucidating the interactions between the pathogenic mechanism of CsCMV and the interactions network of the plant defense system.
