摘要

Abstract

Objective:To investigate the effect of magnesium particles on the phenotypic transformation of macropha-ges and its mechanism.Methods:Live/dead staining and cytoskeleton assay were used to observe the biocompatibility of magnesium particles on macrophages.RT-qPCR,immunofluorescence staining and transcriptome sequencing were used to ob-serve the effect and mechanism of magnesium particles on macrophage phenotype transformation.Results:Macrophages in the magnesium particles group adhered and grew well.Magnesium particles down-regulated the mRNA levels of CCL2,CD86 and IL-1β in inflammatory phenotype(M1 type),and down-regulated the protein level of marker CD86.The mRNA and protein levels of repair phenotype(M2 type)CD206 were up-regulated.Magnesium particles inhibited macrophage inflammation-relat-ed signaling pathways such as tumor necrosis factor production,interleukin-6 production,and NF-κB signaling pathway.Con-clusion:Magnesium particles regulate the formation of anti-inflammatory and pro-repair immune microenvironment by down-regulating the expression of M1 related pathways and genes and up-regulating the expression of M2 related genes in macro-phages.
