Effect of Disaccharides on the Survival Rate of Agrobacterium after Room Temperature Storage

Abstract
To explore the preservation efficacy of disaccharide compounds on Agrobacterium under room temperature conditions.Agrobacterium cells were placed in protectants with different types and concentrations of disaccharide and stored at room temperature to determine their survival rate and ability to infect plants.Agrobacterium survival rate survival rate remains unchanged when stored in a protective agent with a sucrose content of 20%~25%in storage period of 45 days.Survival rate survival rate is higher when the 20%~25%concentration of trehalose used in the protective with a storage period of 45 days for Agrobacterium.In the protective agent containing 20%trehalose and 10%protein,Agrobacterium has a higher survival rate that can be maintained for 45 days,while the transient positive rate of GUS gene expression can still be used for the ability to infect plants.These findings provide insights for enhancing gene high-throughput transformation efficiency and offer valuable references for developing commercial microbial products suitable for ambient-temperature storage.关键词
