Abstract

Cotton is an important economic crop related to the national economy and people's livelihood.China's high-quality cotton production is seriously insufficient,resulting in long-term dependence on imports of high-grade cotton yarn.Therefore,improving fiber quality has become an urgent task in cotton breeding.In this study,an F2 secondary segregating population was constructed using the long-staple introgression line C14 as the male parent and the recurrent parent Jimian 262 as the female parent.Seven candidate regions related to fiber length were obtained by the method of bulked segregant analysis(BSA)combined with next-generation sequencing(NGS).Indel markers were developed in the different regions,and combined with SSR markers,genetic map construction and QTL analysis were performed using JoinMap 4 and WinQTLCart 2.5 software.A total of 3 QTLs related to fiber length were identified,which were qFL-A9-1,qFL-A9-2 and qFL-D13-1,and explaining 11.86%,14.25%and 3.57%of the phenotypic variation,respectively.Among which,qFL-A9-1 and qFL-A9-2 were the two major QTLs with accurate location and high phenotypic variation.The individual genotypes of the InDel marker A9-35 on qFL-A9-1 had very significant differences in fiber length trait,indica-ting this marker could be closely linked to the main QTLs of fiber length on chromosome A9 of upland cotton,which laid a foundation for map-based cloning of cotton fiber length gene,and could be applied to molecular marker-assisted selection breeding and molecular polymerization breeding of cotton fiber quality to improve the efficiency of selection and polymerization.
