淀山湖翘嘴鲌配子冷冻保存技术研究
Study on the cryopreservation technology of gametes of Culter alburnus in Dianshan Lake

Abstract
To protect the fish germplasm resources of Culter alburnus,the cryopreservation protocols for the sperm and oocytes of C.alburnus in Dianshan Lake were optimized.The results showed that the sperm preserved in D-15 as dilution,10%ethylene glycol+5 mg/mL trehalose as cryoprotectant,120 μL of sample loading,and 0.4%NaCl as activation conditions had relative high viability.The sperm cryopreserved for 7 days could be successfully incubated by artificial insemination.There was no significant difference in the embryo development process com-pared with the fresh sperm,indicating that the cryopreserved C.alburnus sperm maintained normal functionality.The highest survival rate of oocytes was preserved in potassium chloride(KCl)buffer as dilution,3.00 mol/L dimethyl sulfoxide+2.00 mol/L propylene glycol+1.00 mmol/L melatonin as vitrification cryoprotectant,which was(63.52±2.17)%.This study established a complete cryopreservation system for the gametes of C.alburnus which can provide technical support for the artificial reproduction,breeding innovation,and germplasm resource conservation of the species.关键词
