大泷六线鱼精子低温冷冻保存方法研究
Cryopreservation methods for sperm in Hexagrammos otakii

Abstract
To establish a cryopreservation protocol for sperm of the fat greenling(Hexagrammos otakii),the effects of extenders,sperm-to-cryoprotectant dilution ratios,and cooling procedures during the freezing process were screened and optimized.Fertilization tests were subsequently conducted using sperm cryopreserved with the opti-mal protocol.The results showed that using fish Ringer's solution supplemented with 20%(v/v)ethylene glycol(EG)as the cryoprotectant led to significantly higher cryopreservation efficiency compared with other groups(P<0.05).The sperm motility and longevity were(69.00±2.65)%and(569.67±20.43)s,respectively.Further-more,when the dilution ratio of sperm to cryoprotectant was 1∶3,the motility of sperm after rewarming and acti-vation was(75.00±1.00)%,which was significantly higher than that in other groups(P<0.05).The sperm had a relatively long lifespan of(731.61±64.23)s,but there was no significant difference in sperm lifespan com-pared with the 1∶1 and 1∶5 dilution ratios(P>0.05).Regarding the cooling procedure,the protocol that in-volved equilibration at 4℃for 30 min,followed by vapor-phase exposure 5 cm above liquid nitrogen for 5 min,and finally plunging into liquid nitrogen resulted in significantly superior post-thaw sperm quality compared to other protocols(P<0.05).Fertilization tests further verified the functionality of the cryopreserved sperm,with the mean hatching rate reaching(56.67±9.29)%.Although it was lower than that of fresh sperm,the difference was not statistically significant(P>0.05).The results indicate that the cryopreservation method for the sperm of H.otakii established in this experiment can effectively maintain sperm motility and fertilization ability,and can be ap-plied to the conservation of germplasm resources and artificial reproduction of the fat greenling.关键词
