摘要

Abstract

Objective To investigate the potential causal association between plasma lipidomics and the risk of osteoar-thritis(OA)using Mendelian randomization(MR)analysis.Methods Summary statistics of genome-wide association studies(GWAS)from a Finnish population,including plasma lipidomic data from 7174 individuals,were used to identify genetic instru-mental variables associated with lipid metabolites.Five MR methods—MR-Egger regression,weighted median,inverse-variance weighted(IVW),simple mode,and weighted mode—were applied,along with Bayesian weighted analysis.Heterogeneity,pleiotro-py,and instrument bias were assessed using Cochran's Q test,MR-Egger intercept test,MR-PRESSO,and leave-one-out sensitivity analysis.Results The IVW analysis identified significant association between eight plasma lipid components and OA risk(P<0.05).No evidence of heterogeneity was detected by Cochran's Q test,andMR-PRESSOanalysis indicated no horizontal pleiotro-py.The leave-one-out analysis confirmed the robustness of individual single nucleotide polymorphisms(SNPs).Conclusion This study reveals a complex causal relationship between specific plasma lipid components and OA risk,providing a valuable insight for future research on lipid-based prevention and treatment strategies for osteoarthritis.
