摘要

Abstract

Objective:To investigate the effect of Baizhu Qiwu Granules on the expression of short-chain fatty acids(SCFAs)and c-Kit in colon tissue of model mice with slow transit constipation(STC),and to explore the relationship be-tween the target of Baizhu Qiwu Granules and SCFAs of intestinal flora.Methods:Forty-two specific pathogen-free(SPF)ICR mice were used in the experiment.They were randomly divided into a control group(n=9)and a modeling group(n=33)using a random number table.An STC mouse model was established by intragastric administration of loperamide hydrochlo-ride for 7 days.After modeling,three mice were randomly selected from each group for model validation.Upon successful validation,the remaining mice(control group:n=6;modeling group:n=30)were used.Mice in the modeling group were fur-ther randomly divided into five groups(n=6 each):the model group,low-dose Baizhu Qiwu Granules group,medium-dose Baizhu Qiwu Granules group,high-dose Baizhu Qiwu Granules group and prucalopride group.The mice in each group were given the corresponding drugs by gavage for 14 days.General signs were observed.Fecal water content and intestinal propul-sion rate were measured.Colon tissue morphology was examined using HE staining,c-Kit expression in colon tissue was de-tected by immunohistochemistry,and SCFAs content in mouse feces was analyzed by gas chromatography-mass spectrometry.Results:Compared with the control group,mice in the model group showed decreased activity,dull and rough fur,slow reac-tion,mental fatigue,reduced fecal output with hard,beaded or pellet-like stools,significantly decreased fecal water content,and a markedly lower intestinal propulsion rate,with the differences being statistically significant(P<0.001).Goblet cells in the colon mucosa were reduced,and c-Kit expression was significantly decreased,with the differences being statistically significant(P<0.01).Compared with the model group,mice in all Baizhu Qiwu Granules dose groups and the prucalopride group showed improved activity and mental state,restored fur gloss,quicker reactions,increased fecal output with soft,rugby ball-shaped stools,significantly increased fecal water content and intestinal propulsion rate,with the differences being statisti-cally significant(P<0.05,P<0.01).The number of goblet cells in colon tissue was restored,c-Kit expression was signifi-cantly increased,and the levels of butyric,hexanoic and valeric acids in feces were elevated,with the differences being sta-tistically significant(P<0.01).Conclusion:Baizhu Qiwu Granules can alleviate symptoms and promote intestinal motility in STC mice.The mechanism may be associated with upregulating c-Kit expression and regulating the intestinal flora metabo-lite SCFAs.
